Revolution® products are compatible with modern infinity corrected microscopes from Leica, Nikon, Olympus or Zeiss to meet your preferred configuration. Note: For laser safety, a 100% side port switch is required. Microscopes need environmental control to maintain living conditions for mammalian live cell preparations. We recommend temperature and CO2 control incubators from Okolab and Tokai Hit. Thermal and mechanical factors can impact the observation of living specimens when image focus is affected by drift or vibration, therefore we recommend that systems are configured on vibration isolation tables.

An important recent innovation in microscope design is focus drift correction (FDC). Focus drift can severely affect time-lapse movies and is especially important in confocal and TIRF imaging, where focus and illumination are tightly constrained. Nikon, Olympus and Zeiss have introduced FDC solutions, which Andor iQ supports to provide long-term drift-free imaging for Revolution® live cell imaging solutions.

Although microscope manufactures produce quality solutions, it is possible to improve speed and convenience of some features with 3rd party devices. One example is the transmitted light (TL) source, which is traditionally a quartz halogen (WH) incandescent lamp. TL provides illumination for phase and DIC, which are used with Live Cell fluorescence measurements to provide “spatial context” by revealing membrane and organelle boundaries. WH lamps not only produce heat, which affects thermal stability, but they are generally shuttered to overcome the slow on and off times required for power supply stabilization. Therefore we recommend replacing these with an LED (white or narrow band) which can be switched on and off in microseconds without any vibration. These properties make it easy to integrate high quality TL imaging within Live Cell fluorescence experiments.

Because we look at the entire system and support many microscopes our products are designed for flexibility. An example is Andor laser combiner (ALC) with the MPU. The CO2 Microscope Stage Incubator is a very compact solution to create a suitable environment for cell culturing right on the microscope stage, allowing to perform prolonged observations of living cells.

In the MSI cells are proven to proliferate as well as in a regular bench-top incubator. Operating temperatures are from 10°C to 50°C and thermal cycles are possible.
